New England Biolabs, Inc. EnGen Cas9 NLS, S. pyogenes – 400 pmol
EnGen Spy Cas9 NLS, is an RNA-guided endonuclease that catalyzes site- specific cleavage of double stranded DNA. The location of the break is within the target sequence 3 bases from the NGG PAM (Protospacer Adjacent Motif) ). The PAM sequence, NGG, must follow the targeted region on the opposite strand of the DNA with respect to the region complementary sgRNA sequence. EnGen Spy Cas9 NLS contains Simian virus 40 (SV40) T antigen nuclear localization sequence (NLS) on the N- and C- termini of the protein.
New England Biolabs, Inc. Blunt/TA Ligase Master Mix – 50 reactions
Blunt/TA Ligase Master Mix is a ready-to-use solution of T4 DNA Ligase, proprietary ligation enhancer, and optimized reaction buffer. This master mix is specifically formulated to improve ligation and transformation of both blunt-end and single-base overhang substrates. The master mix format simplifies reaction set-up, ensures an optimized ratio of enzyme and buffer components, and yields robust, rapid ligation of all types of DNA ends using a short incubation time at room temperature. No thawing is necessary as it remains liquid during storage at -20C. Ligations for subcloning can be carried out in small volumes with low DNA concentrations, allowing users to conserve precious DNA samples and directly transform many strains of chemically competent E. coli without dilution.

A high fidelity restriction endonuclease that recognizes the sequence GC^GGCC_GC. High Fidelity (HF) Restriction Enzymes have 100% activity in rCutSmart Buffer; single-buffer simplicity means more straightforward and streamlined sample processing. HF enzymes also exhibit dramatically reduced star activity. HF enzymes are all Time-Saver qualified and can therefore cut substrate DNA in 5-15 minutes with the flexibility to digest overnight without degradation to DNA. Engineered with performance in mind, HF restriction enzymes are fully active under a broader range of conditions, minimizing off-target products, while offering flexibility in experimental design. NEB extensively performs quality controls on all standard and high-fidelity (HF) restriction enzymes. Examples of nuclease contamination studies for some of our HF restriction enzymes are shown below.

New England Biolabs, Inc. BspQI HF-500 units
A Type IIS restriction endonuclease that recognizes the sequence GCTCTTC(1/4). An E. coli strain that carries the modified cloned BspQI gene from Bacillus sphaericus. BspQI-HF is an engineered and improved version of BspQI. (NEB uses the designation "v2" or "HF" in the name to indicate engineered enzymes.)

Dpn I Restriction Enzyme, is negative for nonspecific nucleases. It is used when cloning into multiple cloning site of pBluescript II phagemid vector. It includes an optimal (10X) buffer and/or a 10X concentrate of Universal buffer.
